I guess that it would be possible that he wanted Rize to become Kanou’s “Kakuhou donor”, but we still need a confirmation for Souta’s motives.
As for the RC cells, it does work on all the senses, but only in ghouls. For the Quinx (who theoretically are 20% ghouls), it seems like all of them have particular good senses, but only one per person: Mutsuki - vision, Saiko and Shirazu - hearing and Urie - smell. After his operation, I assume Urie expected immediate increase of his senses, specially smell, but it did not happen:
I assume their senses became that sharp gradually. With Kaneki was the same.
In the beginning, his senses were pretty much the same as a human’s (he wasn’t even able to tell if Kimi was human or ghoul):
However, his senses became sharper with time:
We have seen that, although a ghoul can heal (Kaneki’s regeneration skills are really high too), sometimes the damage is too much and there can be consequences. For instance, after who-knows-how-many-times Kaneki’s fingers were cut, his nails became black; this is called subungual hematoma (basically, there’s blood under his nails):
So, based on this, it’s safe to say that, after having a centipede in his head for a few days…
…and after having his head stabbed twice through the eyes…
…there were ought to be consequences:
The brain damage caused him amnesia and coordination issues:
The damage to his ear limited his hearing to that of a normal human’s:

And the damage to his eyes created the necessity of using glasses:
